111 Type XVII collagen modulates epidermal patterning

چکیده
Many mammals show epidermal patterning, as seen in alternating mouse tail scales and the microtopography of human skin surface. However, mechanisms underlying development such patterning are poorly understood. Here, we that type XVII collagen (COL17), a niche for stem cells, is factor determining patterning. Col17-null mice exhibit slender scales, condition alleviated by COL17 overexpression. Aberrant cell polarity, feature epidermis, does not account slender-scale phenotype. Skin regeneration after wounding accompanied rescued The revertant mosaicism diseased junctional epidermolysis bullosa demonstrate COL17-positive negative harbor mutually distinct patterns. These results defines scale morphology microtopography. Our study sheds light on role tissue pattern formation.

Type VII and XVII Collagen mRNA Expressions in Regenerated Epidermal Laminae in Chronic Equine Laminitis
To confirm ability forming the basement membrane of the regenerated laminar epidermis (rLE) in chronic laminitis, expression of type VII and type XVII collagen mRNAs in the rLE was studied applying sequences of two type of murine collagens. On northern blot analysis, complement DNA (cDNA) probes adjusted from the murine type VII and type XVII collagen could hybridize with the equine mRNAs, and ...
متن کاملType XVII collagen coordinates proliferation in the interfollicular epidermis
Type XVII collagen (COL17) is a transmembrane protein located at the epidermal basement membrane zone. COL17 deficiency results in premature hair aging phenotypes and in junctional epidermolysis bullosa. Here, we show that COL17 plays a central role in regulating interfollicular epidermis (IFE) proliferation. Loss of COL17 leads to transient IFE hypertrophy in neonatal mice owing to aberrant Wn...
متن کاملTransmembrane Collagen XVII Modulates Integrin Dependent Keratinocyte Migration via PI3K/Rac1 Signaling
The hemidesmosomal transmembrane component collagen XVII (ColXVII) plays an important role in the anchorage of the epidermis to the underlying basement membrane. However, this adhesion protein seems to be also involved in the regulation of keratinocyte migration, since its expression in these cells is strongly elevated during reepithelialization of acute wounds and in the invasive front of squa...
متن کاملCollagen XVII in the Human Brain
Collagens have previously been overlooked for roles in the brain since fibrillar collagens, the best known and most widely studied example of collagens, are not present in the mature central nervous system (CNS). However, over the last decade it has become increasingly apparent that collagens are not merely structural proteins giving strength to tissue, but bio-active molecules with a dynamic r...
متن کاملAnti-idiotypic Antibodies against BP-IgG Prevent Type XVII Collagen Depletion
Bullous pemphigoid (BP) mainly targets type XVII collagen (COL17). Intravenous immunoglobulin (IVIg) is used to treat numerous autoimmune diseases, including BP. The major mechanism of action for IVIG is thought to be its immunomodulatory effect. However, little is known about the precise mechanisms of IVIg in BP. We investigate the cellular effects of IVIg, toward treatments for BP. Keratinocy...
